    Uniform Sampling for Networks of Automata

    We call network of automata a family of partially synchronised automata, i.e. a family of deterministic automata which are synchronised via shared letters, and evolve independently otherwise. We address the problem of uniform random sampling of words recognised by a network of automata. To that purpose, we define the reduced automaton of the model, which involves only the product of the synchronised part of the component automata. We provide uniform sampling algorithms which are polynomial with respect to the size of the reduced automaton, greatly improving on the best known algorithms. Our sampling algorithms rely on combinatorial and probabilistic methods and are of three different types: exact, Boltzmann and Parry sampling

    Construire la territorialité à partir d'une infrastructure pour la mobilité ? L'exemple de la ligne des Alpes

    International audienceIn this proposition we analyse the expectations stakeholders develop about territories and the different visions they have about the relationship between planning and mobility, especially concerning the regional railway system and its potential in terms of servicing metropolitan territories. We show, moreover, how the proximity between researchers and territorial stakeholders brings representations and reciprocal expectations to emerge. The proposed communication is based on an interdisciplinary action-research conducted within the pole Innovation and Mountain Territories (Labex ITEM) of the Grenoble-Alpes University in collaboration with local stakeholders (representatives of institutions, economic and associative networks, inhabitants, etc.). The research, ongoing, focuses on the role of the regional railway line Grenoble-Veynes-Gap in the construction of the Grenoble metropolis. We formulate the hypothesis that the threat of dismissing the line is bringing out more clearly the expectations and the needs of peripheral metropolitan territories in the context of the current French territorial reorganization.Afin de réfléchir aux attentes que les acteurs développent par rapport aux territoires, nous analysons les rapports multiples qui s’instaurent entre les différentes visions des acteurs autour de la relation entre territoire et mobilité, en particulier le système ferroviaire régional et son potentiel en terme de desserte des territoires métropolitains. Nous montrons, de plus, comment la proximité entre chercheurs et acteurs du territoire fait émerger les représentations et les attentes réciproques. La communication proposée s’appuie sur une recherche-action interdisciplinaire conduite au sein du Labex Innovation et territoires de montagne de l’Université Grenoble Alpes en collaboration avec des acteurs locaux (représentants des institutions, du tissu économique et associatif, habitants, etc.).     Diabetic cardiomyopathy: effects of fenofibrate and metformin in an experimental model – the Zucker diabetic rat

    <p>Abstract</p> <p>Background</p> <p>Diabetic cardiomyopathy (DCM) contributes to cardiac failure in diabetic patients. It is characterized by excessive lipids accumulation, with increased triacylglycerol (TAG) stores, and fibrosis in left ventricle (LV). The mechanisms responsible are incompletely known and no specific treatment is presently defined. We evaluated the possible usefulness of two molecules promoting lipid oxidation, fenofibrate and metformin, in an experimental model of DCM, the Zucker diabetic rat (ZDF).</p> <p>Methods</p> <p>ZDF and controls (C) rats were studied at 7, 14 and 21 weeks. After an initial study at 7 weeks, ZDF rats received no treatment, metformin or fenofibrate until final studies (at 14 or 21 weeks). C rats received no treatment. Each study comprised measurements of metabolic parameters (plasma glucose, TAG, insulin levels) and sampling of heart for histology and measurements of TAG content and relevant mRNA concentration.</p> <p>Results</p> <p>ZDF rats were insulin-resistant at 7 weeks, type 2 diabetic at 14 weeks and diabetic with insulin deficiency at 21 weeks. Their plasma TAG levels were increased. ZDF rats had at 7 weeks an increased LV TAG content with some fibrosis. LV TAG content increased in untreated ZDF rats at 14 and 21 weeks and was always higher than in C. Fibrosis increased also moderately in untreated ZDF rats. Metformin and fenofibrate decreased plasma TAG concentrations. LV TAG content was decreased by metformin (14 and 21 weeks) and by fenofibrate (14 weeks). Fibrosis was reduced by fenofibrate only and was increased by metformin. Among the mRNA measured, fenofibrate increased Acyl-CoA Oxidase mRNA level, metformin decreased Acyl-CoA Synthase and increased AdipoR1 and pro-inflammatory mRNA levels.</p> <p>Conclusion</p> <p>Fenofibrate had favourable actions on DCM. Metformin had beneficial effect on TAG content but not on fibrosis. PPARα agonists could be useful for the prevention and treatment of DCM.</p

    Beyond Admissibility: Dominance Between Chains of Strategies

    Admissible strategies, i.e. those that are not dominated by any other strategy, are a typical rationality notion in game theory. In many classes of games this is justified by results showing that any strategy is admissible or dominated by an admissible strategy. However, in games played on finite graphs with quantitative objectives (as used for reactive synthesis), this is not the case. We consider increasing chains of strategies instead to recover a satisfactory rationality notion based on dominance in such games. We start with some order-theoretic considerations establishing sufficient criteria for this to work. We then turn our attention to generalised safety/reachability games as a particular application. We propose the notion of maximal uniform chain as the desired dominance-based rationality concept in these games. Decidability of some fundamental questions about uniform chains is established

    Entrainment, Diffusion and Effective Compressibility in a Self-Similar Turbulent Jet

    An experimental Lagrangian study based on particle tracking velocimetry has been completed in an incompressible turbulent round water jet freely spreading into water. The jet is seeded with tracers only through the nozzle: inhomogeneous seeding called nozzle seeding. The Lagrangian flow tagged by these tracers therefore does not contain any contribution from particles entrained into the jet from the quiescent surrounding fluid. The mean velocity field of the nozzle seeded flow, ⟨Uφ⟩, is found to be essentially indistinguishable from the global mean velocity field of the jet, ⟨U⟩, for the axial velocity while significant deviations are found for the radial velocity. This results in an effective compressibility of the nozzle seeded flow for which ∇⋅⟨Uφ⟩≠0 even though the global background flow is fully incompressible. By using mass conservation and self-similarity, we quantitatively explain the modified radial velocity profile and analytically express the missing contribution associated to entrained fluid particles. By considering a classical advection-diffusion description, we explicitly connect turbulent diffusion of mass (through the turbulent diffusivity KT) and momentum (through the turbulent viscosity νT) to entrainment. This results in new practical relations to experimentally determine the non-uniform spatial profiles of KT and νT (and hence of the turbulent Prandtl number σT=νT/KT) from simple measurements of the mean tracer concentration and axial velocity profiles. Overall, the proposed approach based on nozzle seeded flow gives new experimental and theoretical elements for a better comprehension of turbulent diffusion and entrainment in turbulent jets

    Lagrangian Diffusion Properties of a Free Shear Turbulent Jet

    A Lagrangian experimental study of an axisymmetric turbulent water jet is performed to investigate the highly anisotropic and inhomogeneous flow field. Measurements are conducted within a Lagrangian exploration module, an icosahedron apparatus, to facilitate optical access of three cameras. Stereoscopic particle tracking velocimetry results in three-component tracks of position, velocity and acceleration of the tracer particles within the vertically oriented jet with a Taylor-based Reynolds number Reλ≃230. Analysis is performed at seven locations from 15 diameters up to 45 diameters downstream. Eulerian analysis is first carried out to obtain critical parameters of the jet and relevant scales, namely the Kolmogorov and large (integral) scales as well as the energy dissipation rate. Lagrangian statistical analysis is then performed on velocity components stationarised following methods inspired by Batchelor (J. Fluid Mech., vol. 3, 1957, pp. 67–80), which aim to extend stationary Lagrangian theory of turbulent diffusion by Taylor to the case of self-similar flows. The evolution of typical Lagrangian scaling parameters as a function of the developing jet is explored and results show validation of the proposed stationarisation. The universal scaling constant C0 (for the Lagrangian second-order structure function), as well as Eulerian and Lagrangian integral time scales, are discussed in this context. Constant C0 is found to converge to a constant value (of the order of C0=3) within 30 diameters downstream of the nozzle. Finally, the occurrence of finite particle size effects is investigated through consideration of acceleration-dependent quantities

    Inhibition of the Proprotein Convertases Represses the Invasiveness of Human Primary Melanoma Cells with Altered p53, CDKN2A and N-Ras Genes

    Altered tumor suppressor p53 and/or CDKN2A as well as Ras genes are frequently found in primary and metastatic melanomas. These alterations were found to be responsible for acquisition of invasive and metastatic potential through their defective regulatory control of metalloproteinases and urokinase genes.Using primary human melanoma M10 cells with altered p53, CDKN2A and N-Ras genes, we found that inhibition of the proprotein convertases (PCs), enzymes involved in the proteolytic activation of various cancer-related protein precursors resulted in significantly reduced invasiveness. Analysis of M10 cells and their gastric and lymph node derived metastatic cells revealed the presence of all the PCs found in the secretory pathway. Expression of the general PCs inhibitor alpha1-PDX in these cells in a stable manner (M10/PDX) had no effect on the mRNA expression levels of these PCs. Whereas, in vitro digestion assays and cell transfection experiments, revealed that M10/PDX cells display reduced PCs activity and are unable to process the PCs substrates proIGF-1R and proPDGF-A. These cells showed reduced migration and invasion that paralleled decreased gelatinase MMP-2 activity and increased expression and secretion of tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2. Furthermore, these cells showed decreased levels of urokinase-type plasminogen activator receptor (uPAR) and increased levels of plasminogen activator inhibitor-1 (PAI-1).Taken together, these data suggest that inhibition of PCs activity results in decreased invasiveness of primary human melanoma cells despite their altered p53, CDKN2A and N-Ras genes, suggesting that PCs may serve as novel therapeutic targets in melanoma

    Approximating values of generalized-reachability stochastic games

    Simple stochastic games are turn-based 2½-player games with a reachability objective. The basic question asks whether one player can ensure reaching a given target with at least a given probability. A natural extension is games with a conjunction of such conditions as objective. Despite a plethora of recent results on the analysis of systems with multiple objectives, the decidability of this basic problem remains open. In this paper, we present an algorithm approximating the Pareto frontier of the achievable values to a given precision. Moreover, it is an anytime algorithm, meaning it can be stopped at any time returning the current approximation and its error bound

    Measuring the quality of life of family carers of people with dementia: development and validation of C-DEMQOL

    Purpose We aimed to address gaps identified in the evidence base and instruments available to measure the quality of life (QOL) of family carers of people with dementia, and develop a new brief, reliable, condition-specific instrument. Methods We generated measurable domains and indicators of carer QOL from systematic literature reviews and qualitative interviews with 32 family carers and 9 support staff, and two focus groups with 6 carers and 5 staff. Statements with five tailored response options, presenting variation on the QOL continuum, were piloted (n = 25), pre-tested (n = 122) and field-tested (n = 300) in individual interviews with family carers from North London and Sussex. The best 30 questions formed the C-DEMQOL questionnaire, which was evaluated for usability, face and construct validity, reliability and convergent/discriminant validity using a range of validation measures. Results C-DEMQOL was received positively by the carers. Factor analysis confirmed that C-DEMQOL sum scores are reliable in measuring overall QOL (ω = 0.97) and its five subdomains: ‘meeting personal needs’ (ω = 0.95); ‘carer wellbeing’ (ω = 0.91); ‘carer-patient relationship’ (ω = 0.82); ‘confidence in the future’ (ω = 0.90) and ‘feeling supported’ (ω = 0.85). The overall QOL and domain scores show the expected pattern of convergent and discriminant relationships with established measures of carer mental health, activities and dementia severity and symptoms. Conclusions The robust psychometric properties support the use of C-DEMQOL in evaluation of overall and domain-specific carer QOL; replications in independent samples and studies of responsiveness would be of value
